RNF31 also known as HOIP (HOIL-1-interacting protein) is a part of the linear ubiquitin chain assembly complex (LUBAC). It has a mass of approximately 119 kDa and plays an essential role in protein ubiquitination processes. This protein is actively expressed in various tissues including the spleen lung and lymphoid tissues. RNF31 collaborates with other proteins like SHARPIN and RBCK1 to modulate cellular responses.
Biological function summary

RNF31 contributes to the regulation of NF-κB signaling which is an important player in inflammation and immune responses. It is an important component of the LUBAC complex facilitating the linear ubiquitination of target proteins. Its activity influences the activation of NF-κB transcription factors which are important for controlling the expression of genes linked to immune and inflammatory reactions.

Pathways

The RNF31 protein is integral to the NF-κB signaling pathway as well as to apoptosis. It mediates modifications on proteins that ultimately result in NF-κB activation maintaining a balance between survival and cell death. In these functions RNF31 is closely related to SUV39H1 and other partners within these pathways which collectively influence transcriptional activity and cellular fate.

RNF31 has connections with inflammatory diseases and certain types of cancer. Abnormal regulation or expression of RNF31 and its associated proteins like SHARPIN might contribute to pathological conditions by disrupting normal immune responses. Understanding these connections offers potential therapeutic targets for conditions such as rheumatoid arthritis and various carcinomas.
