REACH authorisation
Abcam has not and does not intend to apply for the REACH Authorisation of customers' uses of products that contain European Authorisation list (Annex XIV) substances.
It is the responsibility of our customers to check the necessity of application of REACH Authorisation, and any other relevant authorisations, for their intended uses.

Lysate preparation: Our lysates are made using RIPA buffer to which we add a protease inhibitor cocktail and phosphatase inhibitor cocktail (ratio: 300:100:10). This means that the protein of interest is denatured. If you require a native form of the protein please use the live cell version. Please refer to our lysis protocol for further details on how our lysates are prepared.

User storage instructions: Lyophilizate may be stored at 4°C. After reconstitution, store at -20°C for short-term storage or -80°C for long-term storage.

This product is subject to limited use licenses from The Broad Institute and ERS Genomics Limited, and is developed with patented technology. For full details of the limited use licenses and relevant patents please refer to our limited use license and patent pages.

Antizyme inhibitor 1 (AZIN1) also known as ornithine decarboxylase antizyme inhibitor (OAZI) is a regulatory protein that modulates polyamine levels in the cell. The mass of AZIN1 is approximately 50 kDa. It is a cytoplasmic enzyme and expressed in various tissues including the liver and pancreas. AZIN1 competes with ornithine decarboxylase (ODC) for binding to antizyme which is a negative regulator of ODC. Unlike ODC AZIN1 lacks catalytic activity but plays an important role in cellular regulation.
Biological function summary

AZIN1 significantly influences polyamine biosynthesis by preventing the formation of inhibitory antizyme-ODC complexes. This effectively regulates polyamine levels that are important for cell growth and proliferation. AZIN1 also interacts with other proteins such as ODC forming a regulatory system to balance polyamine homeostasis. The dynamic interaction within this system is critical for cell function affecting several metabolic processes and cellular activities.

Pathways

AZIN1 integrates importantly into the polyamine biosynthesis pathway. This pathway is essential for providing necessary polyamines like putrescine spermidine and spermine central for cellular replication and differentiation. AZIN1's activity can indirectly influence related signaling pathways through polyamine availability impacting proteins like c-Myc which is known to play roles in cell cycle regulation. The control of polyamine metabolism by AZIN1 reflects its broader role in cellular signaling networks.

AZIN1 exhibits associations with cancer and liver fibrosis. Abnormal expression or mutations in AZIN1 impact liver disease progression by mediating fibrosis-associated pathways. Furthermore AZIN1's involvement in regulating polyamine levels has links to oncogenesis with overexpressed AZIN1 promoting tumor development and progression. AZIN1's interaction with antizyme and the disruption of this interaction can contribute to pathological conditions by impairing normal cellular regulatory mechanisms.
