Interpolated concentrations of native Axl in human serum, and plasma samples.

The concentrations of Axl were measured in duplicates, interpolated from the Axl standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1%, plasma (citrate) 2%, plasma (EDTA) 0.5% and plasma (heparin) 1%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Axl concentration was determined to be 66 ng/mL in serum, 56 ng/mL in plasma (citrate) and 66 ng/mL in plasma (EDTA) and 64 ng/mL in plasma (heparin).

Serum from ten individual healthy human female donors was measured in duplicate.

Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Axl concentration was determined to be 58.1 ng/mL with a range of 37.8 – 89.9 ng/mL.

The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean Axl concentration was determined to be 271 ng/mL in urine and 2.45 ng/mL in milk.

Human Axl ELISA Kit (ab208987) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Axl protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, milk, saliva, serum, and urine. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human Axl with 11 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Axl is a transmembrane receptor tyrosine kinase on the plasma membrane. The growth factor GAS6 is a ligand for Axl and regulates many physiological processes including cell survival, cell proliferation, migration and differentiation. The antibodies used in this kit were raised against a portion of the extracellular domain of Axl.

The Axl protein also known as AXL receptor tyrosine kinase plays a significant role in cell signaling. It has a molecular weight of approximately 98 kDa. Axl is a transmembrane receptor that is expressed in a variety of tissues including the immune system reproductive organs and the central nervous system. It is mainly recognized for transmitting signals from the extracellular matrix into the cytoplasm by binding with its ligand Gas6.
Biological function summary

Axl is important in mediating cell survival proliferation and migration. It functions as part of the TAM family of receptors which also includes Tyro3 and Mer. Receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding growth factor GAS6 and which is thus regulating many physiological processes including cell survival, cell proliferation, migration and differentiation. Ligand binding at the cell surface induces dimerization and autophosphorylation of AXL. Following activation by ligand, AXL binds and induces tyrosine phosphorylation of PI3-kinase subunits PIK3R1, PIK3R2 and PIK3R3; but also GRB2, PLCG1, LCK and PTPN11. Other downstream substrate candidates for AXL are CBL, NCK2, SOCS1 and TNS2. Recruitment of GRB2 and phosphatidylinositol 3 kinase regulatory subunits by AXL leads to the downstream activation of the AKT kinase. GAS6/AXL signaling plays a role in various processes such as endothelial cell survival during acidification by preventing apoptosis, optimal cytokine signaling during human natural killer cell development, hepatic regeneration, gonadotropin-releasing hormone neuron survival and migration, platelet activation, or regulation of thrombotic responses. Also plays an important role in inhibition of Toll-like receptors (TLRs)-mediated innate immune response..
