Abcam's EGF Human ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human EGF in serum, plasma and cell culture supernatants.

This assay employs an antibody specific for Human EGF coated on a 96-well plate. Standards and samples are pipetted into the wells and EGF present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human EGF antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of EGF bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Optimisation may be required with urine samples.

Epidermal Growth Factor (EGF) also known as urogastrone is a single polypeptide that facilitates cell growth proliferation and differentiation by binding to the Epidermal Growth Factor Receptor (EGFR). The EGF protein is characterized by a relatively small molecular weight of approximately 6.4 kDa. It is expressed in various tissues including the kidney submandibular glands and Brunner's glands in the mouse and other species. The presence of anti-EGF antibodies can further enhance the understanding of its mechanical actions and functions in different organisms.
Biological function summary

The epidermal growth factor plays a pivotal role in the regulation of cell signaling. It functions outside a complex interacting mainly with EGFR located on the cell surface. Upon ligand binding EGF induces the receptor dimerization leading to autophosphorylation which activates intrinsic intracellular signaling cascades. This activation is critical for cellular responses such as DNA synthesis and the progression of the cell cycle.
