Interpolated concentrations of native IGFBP2 in human PBMC cell culture supernatant samples treated for 46 hours with 1.5% PHA-M.

The concentrations of IGFBP2 were measured in duplicates, interpolated from the IGFBP2 standard curves and corrected for sample dilution. Undiluted samples are as follows : PBMC supernatant 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP2 concentration was determined to be 3,034.42 pg/mL in PBMC supernatant.

The concentrations of IGFBP2 were measured in duplicates, interpolated from the IGFBP2 standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5%, plasma (citrate) 5%, plasma (heparin) 10%, and plasma (EDTA) 10%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGFBP2 concentration was determined to be 257.87 ng/mL in serum, 273.81 ng/mL in plasma (citrate), 139.86 ng/ml in plasma (heparin), and 118.43 ng/ml in plasma (EDTA).

How the assay works

Human IGFBP2 SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. Multifunctional protein that plays a critical role in regulating the availability of IGFs such as IGF1 and IGF2 to their receptors and thereby regulates IGF-mediated cellular processes including proliferation, differentiation, and apoptosis in a cell-type specific manner (PubMed : 18563800, PubMed : 38796567). Functions coordinately with receptor protein tyrosine phosphatase beta/PTPRB and the IGF1 receptor to regulate IGF1-mediated signaling by stimulating the phosphorylation of PTEN leading to its inactivation and AKT1 activation (PubMed : 22869525). Plays a positive role in cell migration via interaction with integrin alpha5/ITGA5 through an RGD motif (PubMed : 16569642). Additionally, interaction with ITGA5/ITGB1 enhances the adhesion of endothelial progenitor cells to endothelial cells (PubMed : 26076738). Upon mitochondrial damage, facilitates apoptosis with ITGA5 of podocytes, and then activates the phosphorylation of focal adhesion kinase (FAK)-mediated mitochondrial injury (PubMed : 38796567).
