Interpolated concentrations of native IL-18 in human HeLa, HL-60, and A431 cell extract samples based on a 800 μg/ml extract load. The concentrations of IL-18 were measured in duplicate and interpolated from the IL-18 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-18 concentration was determined to be 1,099 pg/ml in HeLa cell extract, 456 pg/ml in HL-60 cell extract and 567 pg/ml in A431 cell extract.

Serum from nine individual healthy human female donors was measured in duplicate. Interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-18 concentration was determined to be 423 pg/mL with a range of undetectable – 2,852 pg/mL.

Human IL-18 ELISA Kit, Chemiluminescent (ab325627) is a rapid single-wash sandwich 55 min ELISA to measure Human IL-18 in Serum, Citrate Plasma, EDTA Plasma, Heparin Plasma, Cell Culture Supernatant, Urine, Cell Extract with a sensitivity of 2.4 pg/mL.

How the assay works

Human IL-18 ELISA Kit, Chemiluminescent (ab325627) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colormetric ELISAs.

Human IL-18 (ab325627) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection

Pro-inflammatory cytokine primarily involved in epithelial barrier repair, polarized T-helper 1 (Th1) cell and natural killer (NK) cell immune responses (PubMed : 10653850). Upon binding to IL18R1 and IL18RAP, forms a signaling ternary complex which activates NF-kappa-B, triggering synthesis of inflammatory mediators (PubMed : 14528293, PubMed : 25500532, PubMed : 37993714). Synergizes with IL12/interleukin-12 to induce IFNG synthesis from T-helper 1 (Th1) cells and natural killer (NK) cells (PubMed : 10653850). Involved in transduction of inflammation downstream of pyroptosis : its mature form is specifically released in the extracellular milieu by passing through the gasdermin-D (GSDMD) pore (PubMed : 33883744).
