Sandwich ELISA - Human MMP12 ELISA Kit (AB255715)
Interpolated concentrations of native MMP12 in human spleen extract based on a 3000 μg/mL extract load.
The concentrations of MMP12 were measured in duplicate and interpolated from the MMP12 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean MMP12 concentration was determined to be 7556.82 pg/mL in spleen extract.

Human MMP12 ELISA Kit (ab255715) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of MMP12 protein in tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate Human MMP12 with 54.9 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples

A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
MMP12 (Matrix metalloproteinase-12), also known as MME or ME, is an enzyme that in humans is encoded by the MMP12 gene. MMP12 is member of a cluster of MMP genes that localize to chromosome 11q22.2. It is thought that the protein encoded by this gene is cleaved at both ends to yield the active enzyme, but this processing has not been fully described. The enzyme degrades soluble and insoluble elastin. MMP12 may play a role in aneurysm formation and in tissue injury and remodeling. Studies in mice suggest a role in the development of emphysema. Human MMP12 shares 62% and 60% sequence homology with mouse and rat respectively.

Biological function summary
MMP12 plays an important role in tissue remodeling and repair processes. It interacts with other proteases within the extracellular matrix. The degradation of elastin by MMP12 contributes to structural organization and it participates in immune response modulation. As part of the matrix metalloproteinase family MMP12 takes part in the larger proteolytic system which comprises multiple enzymes working in concert.
Pathways
MMP12 is an essential factor in the extracellular matrix degradation pathway. This pathway regulates tissue remodeling and inflammatory response. MMP12 independently breaks down elastin while it interacts with proteins like MMP2 and MMP9 which assist in remodeling processes. It also contributes to the regulation of cytokines that orchestrate immune functions.
