Interpolated concentrations of human Pro-MCH in tissue extract
Interpolated concentration of native Pro-MCH was measured in duplicate at different sample concentrations. Undiluted sample are 200 µg/mL human brain hippocampus extract. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). Sample dilutions are made in 1X Cell Extraction Buffer PTR.

Human Pro-MCH SimpleStep ELISA® kit (ab303757) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of Human Pro-MCH protein in tissue extracts. Quantitate Human Pro-MCH with 100.411 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
-Single-wash protocol reduces assay time to 90 minutes or less
-High sensitivity, specificity and reproducibility from superior antibodies
-Fully validated in biological samples
-96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.

Biological function summary
The pro-MCH precursor undergoes cleavage to generate MCH which plays a significant role in energy balance and feeding behavior. MCH functions are often linked to neuronal populations in the hypothalamus that regulate hunger signals and it is involved in forming neuronal circuits that influence behavior related to food intake. Although pro-MCH itself is not part of a larger complex the derived MCH interacts with its receptors to transmit signals.
Pathways
Melanin-concentrating hormone interacts with energy homeostasis and reward circuits in the brain. It is engaged in the melanocortin pathway which balances energy intake and expenditure. MCH modulates signaling by interacting with receptors such as MCHR1 and MCHR2 influencing food-seeking behavior and energy regulation processes. Additionally its regulation aligns with orexigenic (appetite-increasing) peptides such as neuropeptide Y (NPY) coordinating energy-related responses.
