Interpolated concentrations of native Progranulin in human milk saliva and cell culture supernatant samples.

The concentrations of Progranulin were measured in duplicates interpolated from the Progranulin standard curves and corrected for sample dilution. Undiluted samples are as follows : milk 1 : 160 saliva 5% and OVCAR-3 supernatant 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Progranulin concentration was determined to be 360.2 ng/ml in milk 60.4 ng/ml in saliva and 24.4 ng/ml in OVCAR-3 supernatant.

Undiluted samples are as follows : serum 5% plasma (citrate) 6.25% plasma (heparin) 6.25% and plasma (EDTA) 6.25%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Progranulin concentration was determined to be 60.9 ng/mL in serum 47.8 ng/mL in plasma (citrate) 52.2 ng/ml in plasma (heparin) and 49.8 ng/ml in plasma (EDTA).

How the assay works

Human Progranulin ELISA Kit, Chemiluminescent (ab327046) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Human Progranulin (ab327046) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature

Secreted protein that acts as a key regulator of lysosomal function and as a growth factor involved in inflammation, wound healing and cell proliferation (PubMed : 12526812, PubMed : 18378771, PubMed : 28073925, PubMed : 28453791, PubMed : 28541286). Regulates protein trafficking to lysosomes, and also the activity of lysosomal enzymes (PubMed : 28453791, PubMed : 28541286). Also facilitates the acidification of lysosomes, causing degradation of mature CTSD by CTSB (PubMed : 28073925). In addition, functions as a wound-related growth factor that acts directly on dermal fibroblasts and endothelial cells to promote division, migration and the formation of capillary-like tubule structures (By similarity). Also promotes epithelial cell proliferation by blocking TNF-mediated neutrophil activation preventing release of oxidants and proteases (PubMed : 12526812). Moreover, modulates inflammation in neurons by preserving neurons survival, axonal outgrowth and neuronal integrity (PubMed : 18378771).. Granulin-4. Promotes proliferation of the epithelial cell line A431 in culture.. Granulin-3. Inhibits epithelial cell proliferation and induces epithelial cells to secrete IL-8.. Granulin-7. Stabilizes CTSD through interaction with CTSD leading to maintain its aspartic-type peptidase activity.
