Interpolated concentrations of spiked PTH in Human serum, plasma and cell culture media samples.

The concentrations of PTH were measured in duplicate, interpolated from the PTH standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 50%, plasma (citrate) 50%, plasma (EDTA) 50%, plasma (heparin) 50% and 10F RPMI media 100%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean PTH concentration was determined to be 262.33 pg/mL in serum, 258.51 pg/mL in plasma (citrate) and 264.22 pg/mL in plasma (EDTA), 249.32 pg/mL in plasma (heparin), and 300.25 pg/mL in 10F RPMI media 100%.

How the assay works

Human PTH ELISA Kit, Chemiluminescent (ab326892) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA^® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Human PTH (ab326892) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min

Acts by binding to its receptor, PTH1R, activating G protein-coupled receptor signaling (PubMed : 18375760, PubMed : 35932760).
