Abcam's alpha Macroglobulin Mouse in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of alpha Macroglobulin levels in plasma, serum amd urine.

An alpha Macroglobulin specific antibody has been precoated onto 96-well plates and blocked. Standards or test samples are added to the wells and subsequently an alpha Macroglobulin specific biotinylated detection antibody is added and then followed by washing with wash buffer. Streptavidin-Peroxidase Conjugate is added and unbound conjugates are washed away with wash buffer. TMB is then used to visualize Streptavidin-Peroxidase enzymatic reaction. TMB is catalyzed by Streptavidin-Peroxidase to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of alpha Macroglobulin captured in plate.

The entire kit may be stored at -20°C for long term storage before reconstitution - Avoid repeated freeze-thaw cycles.

Alpha Macroglobulin also known as alpha-2-Macroglobulin (A2M) is a large plasma glycoprotein with a molecular mass of approximately 725 kDa. It functions as a broad-spectrum protease inhibitor capable of trapping a wide range of proteases including serine cysteine aspartic and metalloproteases. A2M possesses a "bait region" that captures these proteases facilitating a conformational change that sequesters them. It is highly expressed in the liver and secreted into the bloodstream maintaining its presence at high levels in plasma.
Biological function summary

Alpha-2-Macroglobulin acts as an inhibitor of various proteases contributing to numerous physiological processes. A2M is not a part of a larger complex working independently to regulate protease activity. Its role in binding and inactivating proteases helps control inflammation and tissue degradation making it important for homeostasis. Is able to inhibit all four classes of proteinases by a unique 'trapping' mechanism. This protein has a peptide stretch, called the 'bait region' which contains specific cleavage sites for different proteinases. When a proteinase cleaves the bait region, a conformational change is induced in the protein which traps the proteinase. The entrapped enzyme remains active against low molecular weight substrates (activity against high molecular weight substrates is greatly reduced). Following cleavage in the bait region a thioester bond is hydrolyzed and mediates the covalent binding of the protein to the proteinase (By similarity).
