Interpolated concentrations of native BAFF in mouse serum and plasma samples.
The concentrations of BAFF were measured in duplicate, interpolated from the BAFF standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 5%, plasma (citrate) 5%, plasma (EDTA) 5% and plasma (heparin) 5%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean BAFF concentration was determined to be 22 ng/mL in serum, 16 ng/mL in plasma (citrate), 20 ng/mL in plasma (EDTA), and 24 ng/mL in plasma (heparin).

Mouse BAFF ELISA Kit (ab216948) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of BAFF protein in cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Mouse BAFF with 7.4 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

B-cell activating factor (BAFF) protein is a 309 amino acid cytokine encoded by the TNFSF13B gene. BAFF has been previously referred to as B Lymphocyte Stimulator (BLys), TNF- and APOL- related leukocyte expressed ligand (TALL-1), and Dendritic cell-derived TNF-like molecule (CD257 antigen). BAFF protein is a member of the tumor necrosis factor (TNF) superfamily of ligands. The BAFF cytokine acts as a potent B-cell activator promoting the proliferation and differentiation of B-cells. It is expressed as a type II transmembrane protein on various cell types including monocytes, dendritic cells, and bone marrow stromal cells. The extracellular portion can be cleaved from the membrane, generating a soluble protein fragment. Inadequate levels of BAFF fail to activate B-cells to produce enough immunoglobulin and lead to immunodeficiency. Excessive levels of BAFF cause abnormally high antibody production, resulting in systemic lupus erythematosus, rheumatoid arthritis, and other autoimmune diseases.

Biological function summary
BAFF plays a significant role in the humoral immune response by enhancing B-cell proliferation and function. It belongs to the tumor necrosis factor (TNF) ligand family and interacts as a homotrimer. The BAFF cytokine can bind to receptors such as BAFF-R BCMA (B-cell maturation antigen) and TACI (transmembrane activator and CAML interactor). These interactions regulate B-cell homeostasis promoting development and differentiation processes important for normal immune responses.
Pathways
BAFF is central to the regulation of immune system signaling particularly in B-cell receptor (BCR) signaling and NF-kB pathway activation. The BAFF-R mediates B-cell survival signals through the NF-kB pathway while TACI and BCMA associations convey additional immune responses regulation. These multiple pathways ensure the maintenance of B-cell populations and their ability to produce antibodies.
