Interpolated concentrations of native C Reactive Protein in mouse serum and plasma samples.

The concentrations of C Reactive Protein were measured in duplicates, interpolated from the C Reactive Protein standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 400, plasma (citrate) 1 : 800, plasma (heparin) 1 : 400, and plasma (EDTA) 1 : 1600. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean C Reactive Protein concentration was determined to be 2854 ng/mL in neat serum, 5469 ng/mL in neat plasma (citrate), 3358 ng/mL in neat plasma (heparin), and 7251 ng/mL in neat plasma (EDTA).

Based on a 200 μg/mL extract load and mouse heart tissue extract based on a 500 μg/mL extract load. The concentrations of C Reactive Protein were measured in duplicate and interpolated from the C Reactive Protein standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean C Reactive Protein concentration was determined to be 7416 pg/mL in mouse liver tissue extract and 5978 pg/mL in mouse heart tissue extract.

How the assay works

Mouse C Reactive Protein SimpleStep ELISA®employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. C Reactive Protein displays several functions associated with host defense: it promotes agglutination, bacterial capsular swelling, phagocytosis and complement fixation through its calcium-dependent binding to phosphorylcholine. C Reactive Protein can interact with DNA and histones and it may scavenge nuclear material released from damaged circulating cells. C Reactive Protein is secreted; it forms a homopentamer pentaxin (or pentraxin) which have a discoid arrangement of 5 non-covalently bound subunits. C Reactive Protein binds 2 calcium ions per subunit. The concentration of C Reactive Protein in plasma increases greatly during acute phase response to tissue injury, infection or other inflammatory stimuli. It is induced by IL1/interleukin-1and IL6//interleukin-6.

C-reactive protein (CRP) also known as CRP protein is an acute-phase protein with a molecular mass of approximately 25 kDa. It is primarily synthesized in the liver in response to cytokines. This protein appears in the blood where it binds to phosphocholine expressed on the surface of dead or dying cells. CRP is often used as a biomarker for inflammation in the body and its levels in human and canine (anti CRP used) blood samples are measured using assays like the CRP ELISA test or CRP ELISA kit.
Biological function summary

CRP plays a significant role in the innate immune system by recognizing foreign pathogens and damaged cells. It does not form part of a larger complex but itself can bind to specific ligands initiating the complement cascade. This activation enhances phagocytosis by macrophages contributing to the clearance of necrotic and apoptotic cells. The CRP protein therefore acts as a fast responder in the presence of inflammation making it a useful indicator for monitoring inflammatory diseases.

Pathways

CRP operates predominantly within the acute-phase response pathway. Its production is upregulated by cytokines such as IL-6 which connects it to inflammation-signaling pathways. CRP also interacts with the complement system bridging innate and adaptive immunity. Besides CRP could indirectly influence pathways involving other inflammatory proteins like serum amyloid A and haptoglobin creating a network of responses important for managing inflammation.

Elevated CRP levels are commonly associated with cardiovascular diseases and rheumatoid arthritis. In cardiovascular diseases CRP acts as a marker for atherosclerosis where the protein levels correlate with disease severity. It connects with other proteins involved in inflammatory processes such as fibrinogen to contribute to disease pathology. In rheumatoid arthritis CRP provides insights into disease activity helping in the assessment of treatment responses.
