Interpolated concentrations of native Endostatin in mouse serum and plasma samples.

The concentrations of Endostatin were measured in duplicates interpolated from the Endostatin standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 1 : 200 plasma (citrate) 1 : 200 plasma (EDTA) 1 : 200 and plasma (heparin) 1 : 200. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Endostatin concentration was determined to be 97.87 ng/mL in serum 72.00 ng/mL in plasma (citrate) 85.87 ng/mL in plasma (EDTA) and 101.37 ng/mL in plasma (heparin).

Undiluted samples are as follows : heart supernatant 1 : 8 and kidney supernatant 1 : 200. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean Endostatin concentration was determined to be 7.60 ng/mL in heart supernatant and 67.42 ng/mL in kidney supernatant.

How the assay works

Mouse Endostatin ELISA Kit, Chemiluminescent (ab326393) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Our SimpleStep Ignite™ ELISA kits use recombinant monoclonal antibodies rigorously validated to ensure the highest level of consistency and reproducibility, improved sensitivity and specificity and ease of scalability and security of supply.

Mouse Endostatin (ab326393) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection

Probably plays a major role in determining the retinal structure as well as in the closure of the neural tube.. Non-collagenous domain 1. May regulate extracellular matrix-dependent motility and morphogenesis of endothelial and non-endothelial cells; the function requires homotrimerization and implicates MAPK signaling.. Endostatin. Potently inhibits endothelial cell proliferation and angiogenesis. May inhibit angiogenesis by binding to the heparan sulfate proteoglycans involved in growth factor signaling (PubMed : 9008168). Inhibits VEGFA isoform VEGF165-induced endothelial cell proliferation and migration. Seems to inhibit VEGFA-mediated signaling by blocking the interaction of VEGFA to its receptor KDR/VEGFR2 (PubMed : 12029087). Modulates endothelial cell migration in an integrin-dependent manner implicating integrin ITGA5 : ITGB1 and to a lesser extent ITGAV : ITGB3 and ITGAV : ITGB5 (PubMed : 11158588). May negatively regulate the activity of homotrimeric non-collagenous domain 1 (By similarity).
