Interpolated concentrations of native NT-ProANP in mouse serum and plasma samples.

The concentrations of NT-ProANP were measured in duplicates interpolated from the target standard curves and corrected for sample dilution. Undiluted samples are as follows : serum 25% plasma (citrate) 10% plasma (EDTA) 25% and plasma (heparin) 12.5%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean target concentration was determined to be 5.5 ng/mL in neat serum 40.7 ng/mL in neat plasma (citrate) 24.1 ng/mL in neat plasma (EDTA) and 52.2 ng/mL in neat plasma (heparin).

The concentrations of NT-ProANP were measured in duplicate interpolated from the NT-ProANP standard curves and corrected for sample dilution. Undiluted samples are as follows : mouse heart tissue culture supernatant 1%. The interpolated dilution factor corrected values are plotted (mean +/- SD n=2). The mean target concentration was determined to be 335.7 ng/mL in neat mouse heart tissue culture supernatant. No detectable signal was seen in mouse brain tissue culture supernatant samples (not shown).

How the assay works

Mouse NT-ProANP ELISA Kit, Chemiluminescent (ab327068) SimpleStep Ignite™ ELISA employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step during a 30 min only incubation, significantly reducing assay time. By utilizing a chemiluminscent detection method this improves the sensitivity and dynamic range when compared to traditional colorimetric ELISAs.

Mouse NT-ProANP (ab327068) protocol summary

1. Mix : add samples / standards to the wells together with the capture and detector antibody cocktail. Incubate 30 min at room temperature
2. Wash
3. Add ChemiHRP development solution - incubate for 2 min
4. Read the results on a plate reader capable of luminescence detection

Atrial natriuretic peptide. Hormone that plays a key role in mediating cardio-renal homeostasis, and is involved in vascular remodeling and regulating energy metabolism (PubMed : 12890708, PubMed : 22437503, PubMed : 8760210). Acts by specifically binding and stimulating NPR1 to produce cGMP, which in turn activates effector proteins, such as PRKG1, that drive various biological responses (PubMed : 12890708). Regulates vasodilation, natriuresis, diuresis and aldosterone synthesis and is therefore essential for regulating blood pressure, controlling the extracellular fluid volume and maintaining the fluid-electrolyte balance (PubMed : 22437503, PubMed : 8760210). Also involved in inhibiting cardiac remodeling and cardiac hypertrophy by inducing cardiomyocyte apoptosis and attenuating the growth of cardiomyocytes and fibroblasts (By similarity). Plays a role in female pregnancy by promoting trophoblast invasion and spiral artery remodeling in uterus, and thus prevents pregnancy-induced hypertension (PubMed : 22437503). In adipose tissue, acts in various cGMP- and PKG-dependent pathways to regulate lipid metabolism and energy homeostasis (By similarity). This includes up-regulating lipid metabolism and mitochondrial oxygen utilization by activating the AMP-activated protein kinase (AMPK), and increasing energy expenditure by acting via MAPK11 to promote the UCP1-dependent thermogenesis of brown adipose tissue (By similarity). Binds the clearance receptor NPR3 which removes the hormone from circulation (By similarity).. Long-acting natriuretic peptide. May have a role in cardio-renal homeostasis through regulation of natriuresis, diuresis, vasodilation, and inhibiting aldosterone synthesis. In vitro, promotes the production of cGMP and induces vasodilation. May promote natriuresis, at least in part, by enhancing prostaglandin E2 synthesis resulting in the inhibition of renal Na+-K+-ATPase (By similarity). However reports on the involvement of this peptide in mammal blood volume and blood pressure homeostasis are conflicting; according to a report, in vivo it is not sufficient to activate cGMP and does not inhibit collecting duct transport nor effect diuresis and natriuresis (By similarity). Appears to bind to specific receptors that are distinct from the receptors bound by atrial natriuretic peptide and vessel dilator. Possibly enhances protein excretion in urine by decreasing proximal tubular protein reabsorption (By similarity).. Vessel dilator.
