Mouse TYRO3 ELISA Kit is designed for the quantitative determination of TYRO3 in cell culture supernatants, plasma and serum samples.

∆ Note: Mouse TYRO3 concentration is low in normal serum and plasma samples, and may not be detectable in this assay.

This assay employs an antibody specific for mouse TYRO3 coated on a 96-well plate. Standards and samples are pipetted into the wells and TYRO3 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-mouse TYRO3 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of TYRO3 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

TYRO3 also known as Tyrosine-protein kinase receptor TYRO3 is a membrane protein involved in various cellular processes. The protein has a molecular mass of approximately 104 kDa. Receptor tyrosine kinase that transduces signals from the extracellular matrix into the cytoplasm by binding to several ligands including TULP1 or GAS6. Regulates many physiological processes including cell survival, migration and differentiation. Ligand binding at the cell surface induces dimerization and autophosphorylation of TYRO3 on its intracellular domain that provides docking sites for downstream signaling molecules. Following activation by ligand, interacts with PIK3R1 and thereby enhances PI3-kinase activity. Activates the AKT survival pathway, including nuclear translocation of NF-kappa-B and up-regulation of transcription of NF-kappa-B-regulated genes. TYRO3 signaling plays a role in various processes such as neuron protection from excitotoxic injury, platelet aggregation and cytoskeleton reorganization. Also plays an important role in inhibition of Toll-like receptors (TLRs)-mediated innate immune response by activating STAT1, which selectively induces production of suppressors of cytokine signaling SOCS1 and SOCS3.
