Abcam's IgM Pig ELISA kit (ab190537) is an in vitro enzyme-linked immunosorbent assay (ELISA) for the quantitative measurement of IgM in pig serum and plasma.
In this assay the IgM present in samples reacts with the anti-IgM antibodies which have been adsorbed to the surface of polystyrene microtitre wells. After the removal of unbound proteins by washing, anti-IgM antibodies conjugated with horseradish peroxidase (HRP), are added. These enzyme-labeled antibodies form complexes with the previously bound IgM. Following another washing step, the enzyme bound to the immunosorbent is assayed by the addition of a chromogenic substrate, 3,3',5,5'-tetramethylbenzidine (TMB). The quantity of bound enzyme varies directly with the concentration of IgM in the sample tested; thus, the absorbance, at 450 nm, is a measure of the concentration of IgM in the test sample. The quantity of IgM in the test sample can be interpolated from the standard curve constructed from the standards, and corrected for sample dilution.

Biological function summary
IgM acts as the first line of defense during the initial stages of an immune response. It plays an important part in agglutination and neutralization of pathogens forming a complex that alerts other components of the immune system to take action. IgM exists before class-switching to IgG providing immediate protection. It also plays a role in antibody-dependent cellular cytotoxicity. Although not typically a long-term solution its presence is critical for providing initial protection until more specific antibodies like IgG and IgA are produced.

These interactions connect IgM with other proteins like IgG and IgA influencing various immune response pathways.
