Rat IL-1 alpha ELISA Kit is a single-wash 90-min Simplestep used to quantify Rat IL-1 alpha with a sensitivity of 0.205 pg/ml. The assay uses a simple mix-wash-read protocol with just one incubation and one wash step.

Interpolated concentrations of native IL-1 alpha in rat cell culture supernatant sample.

The concentrations of IL-1 alpha were measured in duplicates, interpolated from the IL-1 alpha standard curves and corrected for sample dilution. Undiluted sample was rat spleen cell supernatant 100%. Cells were treated with 100 ng/mL LPS in RPMI culture media containing glutaMAX and 10% fetal bovine serum over 3 days. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IL-1 alpha concentration was determined to be 251.71 pg/mL in LPS treated spleen cell supernatant 100%.

Rat IL-1 alpha ELISA Kit (ab235646) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of IL-1 alpha protein in cell culture supernatant, cit plasma, edta plasma, hep plasma, and serum. It uses our proprietary SimpleStep ELISA® technology. Quantitate Rat IL-1 alpha with 0.205 pg/ml sensitivity.

SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:

- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies

Cytokine constitutively present intracellularly in nearly all resting non-hematopoietic cells that plays an important role in inflammation and bridges the innate and adaptive immune systems. After binding to its receptor IL1R1 together with its accessory protein IL1RAP, forms the high affinity interleukin-1 receptor complex. Signaling involves the recruitment of adapter molecules such as MYD88, IRAK1 or IRAK4. In turn, mediates the activation of NF-kappa-B and the three MAPK pathways p38, p42/p44 and JNK pathways. Within the cell, acts as an alarmin and cell death results in its liberation in the extracellular space after disruption of the cell membrane to induce inflammation and alert the host to injury or damage. In addition to its role as a danger signal, which occurs when the cytokine is passively released by cell necrosis, directly senses DNA damage and acts as signal for genotoxic stress without loss of cell integrity.
