Interpolated concentrations of native SNAP25 in human cerebellum, mouse brain, and rat brain tissue extracts based on a 50 μg/mL extract load.
The concentrations of SNAP25 were measured in duplicate and interpolated from the SNAP25 standard curve.

The concentrations of SNAP25 were measured in duplicate and interpolated from the SNAP25 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean SNAP25 concentration was determined to be 1591.46 pg/mL.

SNAP25 ELISA Kit (ab256394) is a single-wash 90 min sandwich ELISA designed for the quantitative measurement of SNAP25 protein in tissue extracts. It uses our proprietary SimpleStep ELISA® technology. Quantitate SNAP25 with 19 pg/ml sensitivity.
SimpleStep ELISA® technology employs capture antibodies conjugated to an affinity tag that is recognized by the monoclonal antibody used to coat our SimpleStep ELISA® plates. This approach to sandwich ELISA allows the formation of the antibody-analyte sandwich complex in a single step, significantly reducing assay time. See the SimpleStep ELISA® protocol summary in the image section for further details. Our SimpleStep ELISA® technology provides several benefits:
- Single-wash protocol reduces assay time to 90 minutes or less
- High sensitivity, specificity and reproducibility from superior antibodies
- Fully validated in biological samples
- 96-wells plate breakable into 12 x 8 wells strips
A 384-well SimpleStep ELISA® microplate (ab203359) is available to use as an alternative to the 96-well microplate provided with SimpleStep ELISA® kits.
SNAP25, Synaptosomal nerve-associated protein 25, is a t-snare protein involved in docking and exocytosis of vesicles and neurotransmitter release. It docks with its coreceptor on the vesicle, v-snare, and syntaxin to form a quaternary alpha helix that stabilizes the fusion awaiting a calcium-regulated neurotransmitter exocytosis event. Decreased SNAP25 levels are associated with schizophrenia and attention deficit hyperactivity disorder. Human, mouse and rat SNAP25 protein have 100% conserved amino acid sequence.

Biological function summary
SNARE proteins mediate the fusion of transport vesicles with their target membranes. SNAP25 as part of this complex facilitates synaptic-vesicle exocytosis by forming a tight complex with syntaxin and VAMP (also called synaptobrevin). This interaction promotes the merging of vesicle and plasma membranes enabling efficient neurotransmitter release into the synaptic cleft. SNAP25 undergoes dynamic regulation and modification which is important for precise synaptic function.
Pathways
One observes SNAP25's integration in the process of neurotransmitter release specifically in the exocytosis pathway. It associates closely with other SNARE proteins such as syntaxin-1 and VAMP-2 embodying a core element in synaptic signaling mechanisms. The proper functioning of SNAP25 within this pathway assures the controlled release of neurotransmitters influencing synaptic plasticity and communication.
