The protein known as Metalloprotease Three and ATP/GTP Binding Protein-Like 2 or AGBL2 is part of the cytoplasmic carboxypeptidase family. This enzyme functions by removing glutamate residues from tubulin modifying microtubule properties and dynamics. AGBL2 has a molecular mass of around 83 kilodaltons. Researchers have identified its expression in several tissues including the brain liver and heart suggesting a broad role in the human body.
Biological function summary

AGBL2 participates in the regulation of neuronal microtubule stability. It forms a part of the cytosolic complex that influences cellular functions including maintaining the integrity of the cytoskeleton. AGBL2's activity is essential for the post-translational modification of tubulin which affects cell cycle progression and neuronal signaling. By modulating microtubule behavior AGBL2 contributes importantly to processes like intracellular transport and cell differentiation.

Pathways

AGBL2 actively engages in the regulation of the cytoskeleton. It is involved in the tubulin detyrosination/tyrosination cycle and interacts with other proteins like tubulin and MAP (microtubule-associated proteins) to stabilize or destabilize microtubules as needed. AGBL2 also plays roles in influencing the Wnt signaling pathway critical for developmental processes and cellular communication. By doing so it links with other regulatory proteins to modulate cellular responses.

Researchers have connected AGBL2 to neurodegenerative diseases and cancer. Abnormal expression or activity of AGBL2 can lead to disrupted microtubule dynamics causing neurological conditions such as Alzheimer's disease. AGBL2 interacts with proteins like tau whose hyperphosphorylation is a hallmark of Alzheimer's. Moreover in some cancers altered AGBL2 activity may affect cell proliferation by impacting microtubule stability indicating potential therapeutic targets for intervention.

Metallocarboxypeptidase that mediates deglutamylation of tubulin and non-tubulin target proteins. Catalyzes the removal of polyglutamate side chains present on the gamma-carboxyl group of glutamate residues within the C-terminal tail of tubulin protein. Specifically cleaves tubulin long-side-chains, while it is not able to remove the branching point glutamate. Also catalyzes the removal of polyglutamate residues from the carboxy-terminus of non-tubulin proteins such as MYLK.
