Immunocytochemistry/ Immunofluorescence - Alexa Fluor® 647 Anti-Cleaved PARP1 antibody [Y34] (AB237433)
ab237433 staining Cleaved PARP1 in MCF7 cells Staurosporine-treated (1μM, 5 hr). The cells were fixed with 4% formaldehyde (10 min), permeabilized with 0.1% Triton X-100 for 5 minutes and then blocked with 1% BSA/10% normal goat serum/0.3M glycine in 0.1% PBS-Tween for 1h. The cells were then incubated overnight at +4°C with ab237433 at 1/1000 dilution (shown in red) and ab195887, Mouse monoclonal to alpha Tubulin (Alexa Fluor® 488), at 1/250 dilution (shown in green). Nuclear DNA was labelled with DAPI (shown in blue).
Image was taken with a confocal microscope (Leica-Microsystems, TCS SP8).

Overlay histogram showing MCF7 cells treated (red line) or untreated (green line) with 1 μM Staurosporine for 5 hrs and subsequent staining with ab237433. The cells were fixed with 4% formaldehyde (10 min) and then permeabilized with 0.1% PBS-Triton X-100 for 15 min. The cells were then incubated in 1x PBS/10% normal Goat serum to block non-specific protein-protein interactions followed by the antibody (ab237433) (1x 106 in 100 μl at 0.01 μg/ml (1/50000 dilution)) for 30 min at 22°C.
Isotype control antibody (black line) was Rabbit IgG (monoclonal) Alexa Fluor® 647 (ab199093) used at the same concentration and conditions as the primary antibody. Unlabeled sample (blue line) was also used as a control. Acquisition of >5,000 events were collected using a 40 mW Red laser (640nm) and 670/14 bandpass filter.

Biological function summary
The enzymatic function of PARP1 involves the transfer of ADP-ribose units from NAD+ to target proteins a process known as ADP-ribosylation. PARP1 operates as a part of the base excision repair complex essential in DNA repair processes. The cleaved form of PARP1 no longer facilitates DNA repair marking a shift towards apoptosis. When PARP1 is cleaved it indicates caspase activity implying cells are undergoing programmed cell death.
Pathways
Cleaved PARP1 is deeply involved in the apoptosis and DNA damage response pathways. In the apoptosis pathway PARP1 interacts with key proteins like caspase-3 which cleaves PARP during apoptosis. In the DNA damage response PARP1 collaborates with proteins such as XRCC1 facilitating the base excision repair pathway important for fixing single-strand DNA breaks. These pathways highlight the dual role of PARP1 in promoting cell survival through repair and cell death via apoptosis.
