Alpha-cardiac myosin heavy chain also known as alpha myosin or alpha MHC plays an essential role in muscle contraction within the heart. This motor protein has a molecular mass of approximately 223 kDa. It expresses predominantly in cardiac muscle tissue where it functions as a major component of the thick filaments in cardiac muscle cells. The high expression of this protein in the heart ensures effective force generation during systolic contraction.
Biological function summary

Alpha cardiac myosin heavy chain contributes to the cyclical interaction with actin filaments which results in muscle contraction. It forms part of the myosin molecule complex together with its two light chains. The ATPase activity of alpha MHC enables it to convert chemical energy from ATP hydrolysis into mechanical work driving the sliding of actin filaments. This process is important for the maintenance of cardiac muscle contractility and heart function.

Pathways

Alpha cardiac myosin heavy chain functions as a central element in the cardiac muscle contraction pathway. This pathway is essential for the effective pumping of blood by the heart. In this pathway alpha MHC interacts with actin to facilitate cross-bridge cycling which drives muscle contraction. Additionally it relates closely to other myosin heavy chains such as beta-myosin heavy chain which expresses depending on developmental stage or physiological condition ensuring adaptability of cardiac muscle performance.

Alpha cardiac myosin heavy chain has strong links to hypertrophic cardiomyopathy (HCM) and dilated cardiomyopathy (DCM). Mutations in the alpha MHC gene can lead to impaired contractile functions resulting in these hereditary heart conditions. In HCM alpha MHC mutations often contribute to thickening of the heart muscle while in DCM mutations may lead to weakened contractions and enlargement of the heart chambers. This protein also interacts with other sarcomeric proteins like cardiac troponin T which can similarly influence cardiomyopathy development.
