ARHGEF5 also known as Rho guanine nucleotide exchange factor 5 is a critical protein involved in the regulation of Rho GTPases. This protein acts mechanically as a catalyst exchanging GDP for GTP on Rho GTPases which are powerful regulators of the actin cytoskeleton and cell morphology. ARHGEF5 has an approximate molecular mass of 140 kDa and it is expressed in a variety of tissues including the brain heart and lungs among others. In these locations ARHGEF5 plays an essential role in enabling cellular functions that depend on cytoskeletal dynamics.
Biological function summary

ARHGEF5 actively contributes to cellular processes by modulating actin cytoskeleton organization and cell migration. It typically functions as part of signaling complexes that facilitate cellular communication and response to extracellular signals. This modulation is important for processes like cell adhesion movement and growth which are necessary for normal cellular operations and reactions to environmental changes.

Pathways

ARHGEF5 plays an integral role in the Rho signaling pathway and the focal adhesion pathway. These pathways control various cellular processes including cytoskeleton reorganization and cell adhesion both of which are essential for maintaining proper cellular structure and function. Within these pathways ARHGEF5 interacts with other key proteins such as RhoA and Rac1 amplifying its effect on cellular morphology and movement.

ARHGEF5 is associated with cancer and neurological disorders. Aberrant expression or function of ARHGEF5 has been implicated in the progress of different cancer types due to its role in cell migration and invasion. In neurological disorders ARHGEF5 may affect neuronal migration and connectivity. Additionally through its interactions with proteins like PI3K ARHGEF5 may influence cancer cell survival and proliferation further implicating its role in oncogenesis.

타겟 정보

Guanine nucleotide exchange factor which activates Rho GTPases (PubMed : 15601624). Strongly activates RHOA (PubMed : 15601624). Also strongly activates RHOB, weakly activates RHOC and RHOG and shows no effect on RHOD, RHOV, RHOQ or RAC1 (By similarity). Involved in regulation of cell shape and actin cytoskeletal organization (PubMed : 15601624). Plays a role in actin organization by generating a loss of actin stress fibers and the formation of membrane ruffles and filopodia (PubMed : 14662653). Required for SRC-induced podosome formation (By similarity). Involved in positive regulation of immature dendritic cell migration (By similarity).
