Paraffin-embedded human liver tissue stained for ASAHL/PLT using ab238726 at 1/200 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

ASAHL also known as N-acylsphingosine amidohydrolase or PLT is an enzyme that catalyzes the hydrolysis of ceramide into sphingosine and a free fatty acid. The molecular mass of ASAHL is approximately 45 kDa. This enzyme is highly expressed in many tissues including the brain liver kidney and skeletal muscles. Its activity is essential for the metabolism of sphingolipids playing an important role in maintaining cellular lipid balance and signaling.
Biological function summary

ASAHL participates in regulating apoptosis and cell differentiation by controlling levels of bioactive sphingolipids. Although ASAHL operates as a single enzyme and not part of a larger protein complex its action significantly influences intracellular processes by modulating ceramide levels. Ceramide serves as a precursor for signaling molecules and its degradation by ASAHL leads to the generation of sphingosine which is a lipid mediator involved in various cellular responses including those mediated by interleukin pathways.
