Paraffin-embedded human liver tissue stained for ASAHL/PLT using ab238726 at 1/200 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Biological function summary
ASAHL participates in regulating apoptosis and cell differentiation by controlling levels of bioactive sphingolipids. Although ASAHL operates as a single enzyme and not part of a larger protein complex its action significantly influences intracellular processes by modulating ceramide levels. Ceramide serves as a precursor for signaling molecules and its degradation by ASAHL leads to the generation of sphingosine which is a lipid mediator involved in various cellular responses including those mediated by interleukin pathways.
Pathways
ASAHL plays a vital part in the sphingolipid metabolism pathway. This pathway influences numerous cellular processes related to cell growth differentiation and apoptosis. The enzyme's activity is closely related to other proteins such as ceramide synthases which are involved in ceramide biosynthesis and sphingosine kinase which phosphorylates sphingosine to produce sphingosine-1-phosphate. By interacting with these proteins ASAHL helps maintain the balance of sphingolipid metabolites that control cellular homeostasis.
