ASMT also known as acetylserotonin O-methyltransferase or hydroxyindole O-methyltransferase is an enzyme with a molecular weight of approximately 41-45 kDa. It facilitates the transfer of a methyl group from S-adenosylmethionine to N-acetylserotonin producing melatonin. ASMT is widely expressed in the pineal gland but its presence is also noted in other tissues including the retina and gastrointestinal tract. Its activity plays a critical role in synthesizing melatonin a hormone associated with regulating circadian rhythms.
Biological function summary

ASMT contributes to the synthesis of melatonin. It acts as the last enzyme in the melatonin biosynthesis pathway. Melatonin not only regulates the sleep-wake cycle but also influences various physiological processes. While ASMT is not part of a large protein complex its interaction with substrates such as N-acetylserotonin and S-adenosylmethionine highlights its functional relationship within the biosynthetic machinery.

Pathways

ASMT plays a significant role in the serotonin-melatonin pathway. This pathway starts with tryptophan which converts to serotonin and culminates in melatonin synthesis. ASMT collaborates with other enzymes such as tryptophan hydroxylase and serotonin N-acetyltransferase in these pathways. It directly influences circadian rhythm regulation and light-dark cycle adaptation through melatonin production.

ASMT shows a connection to sleep disorders and certain mood disorders like depression. Disruption in melatonin levels often implicates abnormalities in ASMT function. This disruption also associates ASMT with altered serotonin levels implicating its relationship with serotonin receptor-based pathways present in mood disorders. Researchers continue to explore ASMT’s role looking for therapeutic advancements.
