The Avian Influenza A Hemagglutinin (HA) also known as influenza hemagglutinin is a surface glycoprotein with a molecular mass of approximately 63 kilodaltons. HA is expressed on the envelope of the influenza virus where it facilitates viral entry into host cells. This protein binds to sialic acid receptors on the surface of host cells initiating endocytosis and subsequent viral internalization. HA is a critical determinant of host range and antigenicity in avian influenza viruses.
Biological function summary

The avian protein HA plays a central role in the infection cycle of influenza viruses. As part of the viral capsid hemagglutinin undergoes a conformational change upon receptor binding triggering membrane fusion and release of viral RNA into the host cytoplasm. HA is integral to the structure of the virus working alongside neuraminidase which facilitates release of new viral particles. The interaction between hemagglutinin and host receptor is an important step in viral infectivity and immune system evasion.

Pathways

Hemagglutinin interacts closely with cellular and viral pathways influencing viral replication and assembly. HA operates in conjunction with the neuraminidase protein in the viral life cycle particularly in the replication of the genetic material and assembly of the virion. During the entry pathway the low-pH environment in endosomes prompts a change in HA structure leading to fusion of viral and endosomal membranes an important event for viral genome release into the host cytoplasm.

HA of avian influenza is strongly linked to viral pathogenicity and cross-species transmission. Avian influenza A particularly subtypes like H5N1 can cause severe respiratory diseases in birds and humans with HA being an important virulence factor. Mutations in hemagglutinin can alter viral transmissibility and immune escape impacting disease severity. HA also has potential connections with other influenza-related proteins such as matrix protein 2 (M2) which jointly determine viral pathogenicity and host immune responses.
