Paraffin-embedded human appendix tissue stained for Calpain 2 using ab236650 at 1/300 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized tissue using an HRP conjugated SP system.

Calpain 2 also known as CAPN2 or calpain-2 is a heterodimer protease enzyme weighing around 80 kDa. It belongs to the calpain family which requires calcium ions for activation. Calpain 2 expresses in many tissues including the brain muscles and liver indicating its widespread role in cellular processes. The enzyme comprises a large catalytic subunit and a small regulatory subunit each encoded by different genes.
Biological function summary

Calpain-2 plays a significant role in cell signaling cytoskeletal remodeling and apoptosis regulation. It does not typically function in isolation but partners with other molecules forming complexes necessary for precise proteolytic activity.
