CCP1 also known as Cytosolic Carboxypeptidase 1 is an enzymatic protein important in the modulation of microtubule function. This protein is approximately 76 kDa in mass and is mainly expressed in the brain and testes but it can be found in other tissues as well. CCP1 acts by trimming the C-terminal amino acids of tubulin and other substrates impacting their stability and function. This modification plays a role in determining microtubule dynamics which are important for cell shape division and intracellular transport.
Biological function summary

CCP1 contributes to maintaining cellular functions by removing glutamates from the C-terminal of tubulin impacting microtubule behavior. It forms part of the tubulin tyrosination-detyrosination cycle and interacts with other proteins within the cycle to regulate microtubule length and stability. This regulation is essential for the proper operation of neuronal cells and can influence processes like axonal transport and signaling.

Pathways

CCP1's enzymatic activity links to microtubule and cellular cytoskeleton organization pathways. It influences the dynamic instability of microtubules essential in cell division and intracellular traffic. CCP1 interacts with other tubulin-modifying enzymes like CCP5 which together modify microtubule attributes through polyglutamylation and serve roles in cell motility and structural integrity.

CCP1 alterations or mutations have associations with neurodegenerative disorders and certain forms of cancer. Its dysfunction can lead to the progressive degeneration of neurons connected to Ataxia a condition affecting motor control. Additionally CCP1 mutations or disruptions can link to tubulin-binding proteins contributing to unregulated cell proliferation seen in some cancers. Understanding CCP1's role in these conditions can guide therapeutic strategies targeting these proteins and pathways.

타겟 정보

Metallocarboxypeptidase that mediates protein deglutamylation of tubulin and non-tubulin target proteins (PubMed : 22170066, PubMed : 24022482, PubMed : 30420557). Catalyzes the removal of polyglutamate side chains present on the gamma-carboxyl group of glutamate residues within the C-terminal tail of alpha- and beta-tubulin (PubMed : 22170066, PubMed : 24022482, PubMed : 30420557). Specifically cleaves tubulin long-side-chains, while it is not able to remove the branching point glutamate (PubMed : 24022482). Also catalyzes the removal of polyglutamate residues from the carboxy-terminus of alpha-tubulin as well as non-tubulin proteins such as MYLK (PubMed : 22170066). Involved in KLF4 deglutamylation which promotes KLF4 proteasome-mediated degradation, thereby negatively regulating cell pluripotency maintenance and embryogenesis (PubMed : 29593216).
