The Centrosomal Protein 170 (CEP170) also known as KIAA0470 is a significant component of centrosomal structures. This protein weighs around 170 kDa and finds widespread expression in various human tissues. At a cellular level CEP170 localizes to the spindle apparatus during mitosis where it plays a role in microtubule organization. Through its mechanical function CEP170 contributes to maintaining the structural integrity of centrosomes and helps in organizing microtubules.
Biological function summary

CEP170 participates in cell cycle regulation and aids in the proper division of cells. It engages in a complex with other proteins at the centrosome such as CDK5RAP2 and PCM1 to ensure proper microtubule attachment and alignment during mitosis. Its interactions help to maintain centrosome cohesion and spindle formation therefore ensuring accurate chromosome segregation. Failure in these processes can lead to aneuploidy and potentially to cellular malfunction.

Pathways

CEP170 operates within the microtubule organizing center and the cell cycle checkpoint pathways. It interacts closely with proteins like PLK1 which is important for cell cycle progression and spindle formation. Such interactions indicate its role in multiple cellular pathways that orchestrate successful division and prevent genomic instability. CEP170 also plays a part in the regulation of the p53 signaling pathway influencing cellular responses to DNA damage.

CEP170 has associations with certain cancers such as breast cancer and glioblastoma. In these contexts changes in CEP170 expression or malfunction can disrupt normal cell division therefore contributing to tumorigenesis. Its deregulation often accompanies alterations in other proteins like Aurora A kinase which may complicate normal mitotic processes. These associations indicate that CEP170's role in maintaining genomic integrity can impact disease progression and offers potential avenues for targeted therapy.
