Paraffin-embedded human kidney tissue stained for ch TOG with ab236981 at a 1/400 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

HeLa (Human epithelial cell line from cervix adenocarcinoma) cells stained for ch TOG (Green) using ab236981 at a 1/133 dilution in ICC/IF.

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the primary antibody overnight at 4°C. Secondary used is an Alexa-Fluor®488-conjugated Goat Anti-Rabbit IgG (H+L).

Ch-TOG also known as colonic and hepatic tumor overexpressed gene serves as an important player in microtubule dynamics. With a molecular weight of approximately 217 kDa this protein is an important component in microtubule polymerization. It is expressed in various tissues including the brain and testis demonstrating its diverse role in cellular structure and division processes. As an integral part of the microtubule organizing center ch-TOG directly contributes to the assembly and stability of the mitotic spindle.
Biological function summary

Ch-TOG is highly involved in cell cycle regulation and mitosis. By interacting with microtubules it maintains the proper formation of the mitotic spindle ensuring accurate chromosome segregation in eukaryotic cells. It functions within larger protein complexes such as the Augmin complex which assists in spindle microtubule nucleation.
