COQ4 also known as Coenzyme Q4 homologue is a protein that plays an important role in the biosynthesis of coenzyme Q10 (CoQ10) a vital component of the electron transport chain in mitochondria. COQ4 has a molecular mass of approximately 27.5 kDa. It is mainly expressed in tissues with high energy demands like the heart liver and skeletal muscle. COQ4 is important in maintaining mitochondrial function due to its involvement in the production of CoQ10.
Biological function summary

COQ4 associates with the mitochondrial inner membrane and forms part of the multi-subunit complex known as the CoQ biosynthesis complex. This complex is responsible for the stepwise synthesis of CoQ10 which acts as an electron carrier and antioxidant. By participating in this complex COQ4 contributes to cellular respiration and energy production. Its function ensures the stability and assembly of the complex highlighting its essential role in the biochemical pathway of CoQ10 biosynthesis.

Pathways

COQ4 is involved in the oxidative phosphorylation pathway critical for ATP production in eukaryotic cells. It interlinks with the mevalonate pathway as the biosynthesis of CoQ10 shares intermediates derived from this pathway. The protein also works in conjunction with other components of the CoQ biosynthesis complex including COQ3 COQ6 and COQ7 to fulfill its function in mitochondrial electron transport and energy metabolism.

COQ4 mutations and deficiencies have been linked to coenzyme Q10 deficiency syndromes and mitochondrial complex I deficiency. Such conditions can lead to a wide range of symptoms including encephalomyopathy myopathy and organ dysfunction. COQ4's malfunction affects energy production in cells particularly in tissues with high energy requirements. Its connection to other proteins in the CoQ10 biosynthesis pathway such as COQ2 and COQ9 further emphasizes its significance in maintaining cellular energy balance and preventing related disorders.
