CTDSP2 also known as CTD Small Phosphatase 2 plays a role in regulating transcription processes by dephosphorylating the C-terminal domain of RNA polymerase II. This protein has a molecular mass around 33 kDa and is expressed in various tissues including the brain and testis. It possesses phosphoserine and phosphothreonine phosphatase activity which is central to its mechanical function in controlling transcriptional elongation and processing.
Biological function summary

CTDSP2 acts by modulating transcription elongation and RNA processing. It forms part of a complex involving other CTD phosphatases ensuring precise regulation of RNA polymerase II activity. This function implies that CTDSP2 participates in managing the transcription cycle and impacts RNA synthesis critical for gene expression and cellular function.

Pathways

CTDSP2 connects to the RNA polymerase II transcription cycle specifically affecting the transcriptional regulation pathway. It functions alongside associated phosphatases like CTDSP1 and CTDSP3 orchestrating the phosphorylation state of RNA polymerase II. These interactions highlight CTDSP2's integration in the fine-tuning of RNA synthesis impacting gene expression fidelity and cellular responses.

Aberrant activity of CTDSP2 associates with neurodegenerative diseases and cancer. Dysregulation in its expression or function often disrupts cellular transcription equilibrium connecting to proteins such as p53 in cancer pathways where it might influence tumor suppressor activities. Similarly CTDSP2 can impact neurodegenerative diseases through pathways involving amyloid precursor proteins showcasing its implication in brain functionality and pathology.

타겟 정보

Preferentially catalyzes the dephosphorylation of 'Ser-5' within the tandem 7 residue repeats in the C-terminal domain (CTD) of the largest RNA polymerase II subunit POLR2A. Negatively regulates RNA polymerase II transcription, possibly by controlling the transition from initiation/capping to processive transcript elongation. Recruited by REST to neuronal genes that contain RE-1 elements, leading to neuronal gene silencing in non-neuronal cells. May contribute to the development of sarcomas.
