Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-DDHD2 antibody (AB235832)

Immunohistochemical analysis of paraffin-embedded Human gastric cancer tissue labeling DDHD2 with ab235832 at 1/50 dilution followed by a ready to use LeicaDS9800 (Bond™ Polymer Refine Detection). After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a Goat anti-rabbit polymer IgG labeled by HRP and visualized using 0.05% DAB. Secondary antibody only control : uses 1% BSA instead of primary antibody.

DDHD2 also known as PA-PLA1 or KIAA0725 is a phospholipase enzyme with a molecular mass of approximately 90 kDa. This enzyme plays a role in lipid metabolism by hydrolyzing phosphatidic acid. DDHD2 is expressed mainly in the brain suggesting a specialized function in neural tissue. Its enzymatic activity contributes to the dynamic regulation of lipid composition within neuronal membranes influencing membrane fluidity and signaling cascades.
Biological function summary

DDHD2 serves as a critical player in neural lipid homeostasis. It exists as a part of a larger phospholipase complex that regulates various lipid signaling pathways in neurons. The enzyme's activity affects the production of lysophosphatidic acid and other bioactive lipids impacting processes like cell signaling growth and maintenance of neuronal membranes. Loss of DDHD2 activity can therefore disrupt lipid balance affecting neuronal function.

Pathways

DDHD2 actively participates in the phospholipase signaling pathway and the endocannabinoid signaling pathway. Through its interactions DDHD2 influences these pathways by modulating lipid mediators that serve as signaling molecules. It functions along with proteins such as lipin and phospholipase C in these pathways collectively impacting membrane lipid turnover and intracellular signaling events.

DDHD2 is linked with hereditary spastic paraplegia a condition characterized by progressive spasticity and weakness of the lower limbs. Mutations in DDHD2 can lead to defective lipid metabolism in neuronal cells contributing to the pathology of this disorder. Additionally DDHD2 is connected with proteins such as spastin and atlastin which are also implicated in hereditary spastic paraplegia sharing roles in maintaining neuron structure and function.

Diacylglycerol (DAG) and triacylglycerol (TAG) lipase required for proper lipid homeostasis in the central nervous system (PubMed : 29278326, PubMed : 37832604). It cooperates with PNPLA2/ATGL in neuronal TAG catabolism and hydrolyzes sn-1,3 DAG downstream of PNPLA2/ATGL (By similarity). In vitro, it also acts as a phospholipase that hydrolyzes preferentially phosphatidic acids, including 1,2-dioleoyl-sn-phosphatidic acid, phosphatidylcholine and phosphatidylethanolamine. Specifically binds to phosphatidylinositol 3-phosphate (PI(3)P), phosphatidylinositol 4-phosphate (PI(4)P), phosphatidylinositol 5-phosphate (PI(5)P) and possibly phosphatidylinositol 4,5-bisphosphate (PI(4,5)P2). May be involved in the maintenance of the endoplasmic reticulum and/or Golgi structures. May regulate the transport between Golgi apparatus and plasma membrane.
