추가 정보

DOCK6 or Dedicator of Cytokinesis 6 functions as a guanine nucleotide exchange factor (GEF) involved in cytoskeleton organization. This protein weighs approximately 226 kDa. It facilitates the transition between inactive GDP-bound states to active GTP-bound states for small GTPases such as Rac1 and Cdc42. Expression of DOCK6 is widespread but found prominently in neural tissues indicating its relevance in neural development and function.
Biological function summary

DOCK6 acts as an activator of Rho family GTPases which play a central role in various cellular processes including cell migration adhesion and morphology. DOCK6 does not function as part of a larger multi-protein complex but interacts selectively with other proteins to exert its biological effects. The protein is important for proper neuronal network formation influencing the branching and guidance of axons and dendrites.

Pathways

DOCK6 is involved in the Rho GTPase signaling pathway and the cell polarity establishment pathway. In the Rho GTPase signaling pathway DOCK6 interacts closely with proteins such as Rac1 and Cdc42 to modulate cytoskeletal rearrangements. These signaling pathways are key for developmental processes and defects can lead to improper cellular functions impacting tissue architecture and organogenesis.

Mutations in DOCK6 are associated with Adams-Oliver syndrome and microcephaly. Adams-Oliver syndrome which involves limb abnormalities and scalp defects has connections to DOCK6's role in cytoskeleton dynamics. Similarly its link to microcephaly comes from its effects on neuronal differentiation and proliferation. Cdc42 which operates in similar pathways shows overlapping roles in neural development and diseases connected to cytoskeletal disruptions.
