DORFIN also known as RNF19A is an E3 ubiquitin-protein ligase with significant roles in cellular protein degradation. It has a molecular mass of approximately 46 kDa. DORFIN is expressed in several tissues with higher expression noted in the brain and testis. It plays a critical role in the ubiquitin-proteasome system facilitating the tagging of proteins for degradation.

Biological function summary

DORFIN functions by promoting the degradation of misfolded or damaged proteins. It is a component of protein quality control machinery ensuring cellular homeostasis. DORFIN often works in concert with other ubiquitin ligases and proteins including the C-terminus of Hsc70-interacting protein (CHIP) to maintain protein stability. This synergistic action supports cellular stress response and protects against proteotoxic stress.

Pathways

DORFIN participates in the protein ubiquitination and proteasome-mediated degradation pathway. It interacts with and possibly modulates key proteins in the apoptosis pathway such as Parkin. E3 ubiquitin-protein ligase which accepts ubiquitin from E2 ubiquitin-conjugating enzymes UBE2L3 and UBE2L6 in the form of a thioester and then directly transfers the ubiquitin to targeted substrates, such as SNCAIP or CASR. Specifically ubiquitinates pathogenic SOD1 variants, which leads to their proteasomal degradation and to neuronal protection.
