Goat anti-rabbit IgG was coated onto 96-wells. Serial dilution of Estrone-6 (0, 15, 30, 100, 250, 500, 1000, 2000pg/mL) was added (25μL). This was followed by adding of 50μL of HRP-conjugated Estrone and 2.0-2.5ng/mL ab241064 (50μL) into each well. Plates were incubated for 2 hours at room temperature by shaking. After washing, 125μL of Tetramethylbenzidine (TMB) substrate was added and incubated for 30 minutes without shaking for color development. Then 125 μL of stopping solution (acid solution) was dispensed and OD was read at 450nm on a microplate reader within 5 minutes.

Estrone also known as E1 acts as a steroidal estrogen hormone with a molecular weight of approximately 270.4 g/mol. It is commonly expressed in the ovaries adipose tissue and liver. Estrone serves as one of the three major estrogens found in humans alongside estradiol and estriol. As a part of estrogen family it engages in binding to estrogen receptors influencing gene expression and physiological responses in target tissues.
Biological function summary

Estrogen hormones like estrone play significant roles in feminine reproductive health and secondary sexual characteristics. Estrone maintains a balance through conversion to and from other estrogens within the body. It does not function as a complex but influences biological processes such as cell proliferation and tissue growth particularly in reproductive organs. By interacting with cellular mechanisms estrone significantly impacts various physiological responses.

Pathways

Steroid hormones such as estrone integrate into metabolic and reproductive pathways. In the biosynthesis of steroids estrone converts from androstenedione or estradiol through enzymatic actions.
