FBXL18 also known as F-box and leucine-rich repeat protein 18 plays a role as part of the ubiquitin-proteasome system. It has a mass of approximately 60 kDa. Functionally FBXL18 acts as a substrate recognition component of the SCF (SKP1-CUL1-F-box protein) E3 ubiquitin ligase complex. This protein targets specific substrates for ubiquitination marking them for degradation by the proteasome. FBXL18 shows expression in various tissues with notable presence in gastroenterological and neural tissues.
Biological function summary

This protein regulates protein homeostasis and cellular processes by controlling the degradation of target proteins. FBXL18 forms a complex with SKP1 CUL1 and RBX1 in the SCF complex which facilitates the ubiquitination process. Through this function FBXL18 plays a role in processes like cell cycle regulation signal transduction and transcriptional regulation influencing key cellular functions.

Pathways

FBXL18 integrates into the Wnt/β-catenin and NF-κB signaling pathways. In the Wnt/β-catenin pathway FBXL18 interacts with β-catenin playing a part in its degradation and therefore influencing cellular decisions regarding proliferation and differentiation. In the context of the NF-κB pathway FBXL18 affects the stability of proteins like IκB which modulates NF-κB transcription factor activity. This connects FBXL18 to proteins such as β-catenin and IκB within these pathways highlighting its influence on these important signaling cascades.

FBXL18's dysregulation links to cancer and neurodegenerative diseases. Alterations in its expression or function can lead to disrupted control of protein degradation contributing to oncogenesis through aberrant Wnt/β-catenin signaling or affecting neurodegenerative processes by impacting pathways such as NF-κB. In cancer connections to proteins like β-catenin suggest that FBXL18 affects tumor progression and metastasis. Through neurodegenerative diseases FBXL18 interacts with proteins like IκB implicating its role in cellular stress responses and neuronal cell survival.
