GNAT1 also known as Guanine Nucleotide-Binding Protein G(t) Subunit Alpha-1 is an essential component of the visual process. This protein functions as the alpha-subunit of transducin an important player in the phototransduction cascade. GNAT1 has a molecular mass of approximately 40 kDa and is mainly expressed in photoreceptor cells of the retina. It participates in the conversion of photons into electrical signals by activating the cGMP phosphodiesterase enzyme upon stimulation by light-activated rhodopsin.

Biological function summary

GNAT1 plays a fundamental role in vision by mediating signal transduction in rods. It is part of the larger transducin protein complex which includes beta and gamma subunits. Upon activation by light GNAT1 binds to GTP and dissociates from the beta-gamma dimer to interact with downstream effectors initiating a series of reactions that lead to hyperpolarization of the photoreceptor cell membrane.

Pathways

GNAT1 functions within the phototransduction pathway an important system for visual signal processing. This pathway involves key interactions with proteins like rhodopsin and cGMP phosphodiesterase. In the presence of light GNAT1 converts GDP to GTP facilitating a cascade that results in the reduction of cGMP levels which closes cGMP-gated ion channels and alters the membrane potential effectively translating light stimuli into visual signals.

GNAT1 has significant associations with conditions affecting vision such as retinitis pigmentosa. Mutations in the GNAT1 gene can disrupt normal function and lead to defects in the phototransduction pathway contributing to progressive vision loss. Furthermore GNAT1's role in congenital stationary night blindness links it with proteins like rhodopsin and PDE6B emphasizing its involvement in maintaining proper visual function and its interactions that when disrupted result in ocular diseases.

Functions as a signal transducer for the rod photoreceptor RHO. Required for normal RHO-mediated light perception by the retina (PubMed : 22190596). Guanine nucleotide-binding proteins (G proteins) function as transducers downstream of G protein-coupled receptors (GPCRs), such as the photoreceptor RHO. The alpha chain contains the guanine nucleotide binding site and alternates between an active, GTP-bound state and an inactive, GDP-bound state. Activated RHO promotes GDP release and GTP binding. Signaling is mediated via downstream effector proteins, such as cGMP-phosphodiesterase (By similarity).
