The Hantavirus glycoprotein G2 sometimes referred to simply as G2 is an integral membrane protein that plays a critical role in the virus's life cycle. Mechanically G2 facilitates viral entry into host cells by mediating the fusion of the viral envelope with the host cell membrane. This protein is expressed on the surface of the hantavirus envelope and together with glycoprotein G1 forms the glycoprotein complex essential for viral infectivity. The exact molecular mass of the G2 protein can vary between hantavirus strains but typically ranges around 30 kilodaltons.
Biological function summary

The G2 protein plays a pivotal role in the host-pathogen interaction mediating the initial binding and entry process of the virus into host cells. On the surface of the virus the glycoprotein complex consisting of G1 and G2 binds to cellular receptors triggering endocytosis and facilitating the release of viral RNA into the cytoplasm. This complex assembly accounts for the efficient recognition and invasion of permissive cells enabling the virus to hijack host machinery for replication.

Pathways

Hantavirus glycoprotein G2's function integrates into the endocytosis and viral replication pathways. The glycoprotein machinery cooperates with cellular proteins to accomplish membrane fusion and viral internalization. Acidification within endocytic vesicles modulates the conformational change of G2 for membrane fusion linking it indirectly to proteins involved in cellular trafficking such as clathrin and dynamin. These interactions are important for the cytoplasmic release of viral components and subsequent viral propagation.

Hantavirus glycoprotein G2 is closely associated with the pathogenesis of hantavirus infections including hemorrhagic fever with renal syndrome (HFRS) and hantavirus pulmonary syndrome (HPS). The interaction between the G2 protein and host endothelial cells is an important factor in the vascular permeability and endothelial dysfunction observed in these diseases. While G2 interacts directly with the host receptors in the context of these disorders it functions alongside cellular proteins that may impact vascular integrity and immune responses such as integrins and VEGF receptors contributing to the disease's clinical manifestations.
