Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - HRP Anti-DDB1 antibody [EPR6089] (AB199026)
IHC image of DDB1 staining in a section of formalin-fixed paraffin-embedded normal human colon tissue*, performed on a Leica BONDTM. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20mins. The section was then incubated with ab199026, 1/100 dilution, for 15 mins at room temperature. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX. The inset negative control image is taken from an identical assay without primary antibody.
For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Western blot - HRP Anti-DDB1 antibody [EPR6089] (AB199026)
This blot was produced using a 4-12% Bis-tris gel under the MOPS buffer system. The gel was run at 200V for 50 minutes before being transferred onto a Nitrocellulose membrane at 30V for 70 minutes. The membrane was then blocked for an hour using 2% Bovine Serum Albumin before being incubated with ab199026 overnight at 4°C. Antibody binding was visualised using ECL development solution ab133406.

HepG2 (Human hepatocellular liver carcinoma cell line) Whole Cell Lysate at 10 µg
Lane 3:
NIH 3T3 (Mouse embryonic fibroblast cell line) Whole Cell Lysate at 10 µg
Lane 4:
Human placenta tissue lysate - total protein (<a href='/ko/products/unavailable/human-placenta-tissue-lysate-total-protein-ab29745'>ab29745</a>) at 10 µg

Biological function summary
In terms of cellular processes beyond DDB1's immediate interactions it plays a critical role in DNA repair mechanisms and the cell cycle. DDB1 is part of the larger CUL4-DDB1 ubiquitin ligase complex which targets specific proteins for ubiquitination and subsequent degradation therefore facilitating DNA repair and regulating cell cycle progression. This ability to target damaged proteins or signaling errors for removal helps maintain cellular health and preserve genetic information integrity.
Pathways
DDB1 is pivotal in the nucleotide excision repair pathway and the DNA damage response pathway. It interacts dynamically with proteins such as XPC (xeroderma pigmentosum group C) to initiate repair mechanisms ensuring the proper removal of damaged DNA sections. Moreover DDB1's role in ubiquitination connects it to pathways regulating protein turnover and cellular homeostasis interacting with the ubiquitin-proteasome system which is important for controlling protein degradation.
