Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections) - Anti-HYPE antibody (AB237521)

Paraffin-embedded human kidney tissue stained for HYPE using ab237521 at 1/500 dilution in immunohistochemical analysis.

After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

Immunocytochemistry/ Immunofluorescence - Anti-HYPE antibody (AB237521)

PC-3 (Human prostate adenocarcinoma cell line) cells stained for HYPE (green) using ab237521 at 1/166 dilution in ICC/IF. Secondary antibody is an Alexa Fluor® 488-conjugated Goat Anti-Rabbit IgG (H+L). The nuclear counter stain is DAPI (blue).

The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C.

추가 정보

HYPE also known as RHBDF1 is an endoplasmic reticulum protein that functions as a membrane protein involved in protein regulation and trafficking. It has an approximate mass of 38 kDa. HYPE is expressed widely within various tissues but shows significant levels in cells of the liver and kidney. This protein plays a role in multiple cellular processes reflecting its versatile functionality.
Biological function summary

This protein impacts endoplasmic reticulum stress responses and protein homeostasis. It does not seem to form part of a larger complex but operates independently to regulate the degradation of specific proteins aiding in maintaining cellular balance. HYPE's actions contribute to preventing protein aggregation that could disrupt cellular function.

Pathways

HYPE participates in the unfolded protein response (UPR) and the ER-associated degradation (ERAD) pathway. It acts alongside proteins involved in protein folding and trafficking such as BiP and calnexin to ensure synthesis and processing occur efficiently. These pathways highlight HYPE’s interaction with mechanisms that manage protein quality control.

This protein has connections to liver fibrosis and kidney dysfunction. Its regulation of protein folding is significant in these diseases where improper protein management can lead to cellular damage. Researchers have identified its interaction with proteins like TGF-beta in the context of fibrosis linking its importance to disease progression and providing potential therapeutic targets.

타겟 정보

Protein that can both mediate the addition of adenosine 5'-monophosphate (AMP) to specific residues of target proteins (AMPylation), and the removal of the same modification from target proteins (de-AMPylation), depending on the context (By similarity). The side chain of Glu-231 determines which of the two opposing activities (AMPylase or de-AMPylase) will take place (PubMed : 36136088). Acts as a key regulator of the ERN1/IRE1-mediated unfolded protein response (UPR) by mediating AMPylation or de-AMPylation of HSPA5/BiP (PubMed : 25601083, PubMed : 36136088). In unstressed cells, acts as an adenylyltransferase by mediating AMPylation of HSPA5/BiP at 'Thr-518', thereby inactivating it (By similarity). In response to endoplasmic reticulum stress, acts as a phosphodiesterase by mediating removal of ATP (de-AMPylation) from HSPA5/BiP at 'Thr-518', leading to restore HSPA5/BiP activity (By similarity). Although it is able to AMPylate RhoA, Rac and Cdc42 Rho GTPases in vitro, Rho GTPases do not constitute physiological substrates (PubMed : 19362538, PubMed : 25601083).
