IPS1 was immunoprecipitated from HEK-293T (human epithelial cell line from embryonic kidney transformed with large T antigen) whole cell lysate (1.0 mg per IP reaction; 20% of IP loaded) prepared using NETN lysis buffer, using ab240581 at 6 μg per reaction. For western blotting, ab240581 was used at 0.4 μg/ml.

Lane 1 : ab240581 used for IP at 6 μg per reaction.
Lane 2 : Control IgG.

Detection : Chemiluminescence with an exposure time of 30 seconds.

IPS1 also known as MAVS (Mitochondrial Antiviral-Signalling protein) functions at the mechanical level as an important adaptor protein in antiviral immune responses. It exhibits a mass of approximately 56 kDa and is predominantly expressed in the mitochondria of various cells especially immune cells. IPS1 interacts with downstream signaling molecules to execute its functions. It plays an important role by acting as a bridge that links cytosolic sensors of viral RNA to the activation of transcription factors.
Biological function summary

IPS1 mediates the innate immune response against viruses by facilitating the production of type I interferons and inflammatory cytokines. Upon detection of viral components IPS1 forms signalosome complexes with proteins like RIG-I and MDA5 that sense viral RNA. These interactions enable the recruitment of downstream effectors that activate transcription factors such as IRF3 IRF7 and NF-kB leading to an antiviral state. The ability of IPS1 to form these complexes makes it a central modulator in the immune signaling cascade.

Pathways

Different signaling events rely on IPS1 for the propagation of immune signals. IPS1 operates within the RIG-I-like receptor (RLR) pathway an essential component of innate antivirus defense. It plays a pivotal role in signaling cascades that start from RIG-I and MDA5 leading to the activation of IRF and NF-kB transcription factors. These interactions highlight IPS1's integration in pathways that orchestrate cellular responses to viral infection.

IPS1 connects significantly to viral infections and related immune dysfunctions. Abnormal IPS1 signaling can contribute to the pathogenesis of diseases such as hepatitis and systemic lupus erythematosus (SLE). For instance defective interactions between IPS1 and proteins like RIG-I or MDA5 can impair the proper immune response to viral infections leading to chronic infection or inflammatory disorders. Additionally its involvement in triggering inappropriate immune responses further links it to autoimmune conditions such as SLE.

Bifunctional inositol kinase that acts in concert with the IP6K kinases IP6K1, IP6K2 and IP6K3 to synthesize the diphosphate group-containing inositol pyrophosphates diphosphoinositol pentakisphosphate, PP-InsP5, and bis-diphosphoinositol tetrakisphosphate, (PP)2-InsP4. PP-InsP5 and (PP)2-InsP4, also respectively called InsP7 and InsP8, regulate a variety of cellular processes, including apoptosis, vesicle trafficking, cytoskeletal dynamics, exocytosis, insulin signaling and neutrophil activation.
