The KCNA5 protein also known as KV1.5 is a voltage-gated potassium channel. It has a molecular mass of approximately 70 kDa. KCNA5 facilitates potassium ion flow across cardiac and neuronal cell membranes. This protein predominantly expresses in the heart and the central nervous system. It plays a significant role in repolarizing cardiac action potentials contributing to cardiac rhythm regulation.
Biological function summary

KCNA5 functions as part of the KV1 family of potassium channels. It contributes to determining the duration of the action potential by mediating the delayed rectifier current. KCNA5 often forms tetrameric complexes which consist of four KCNA5 subunits or sometimes it forms heteromultimers with other potassium channels. This ability to form functional complexes allows it to modulate cellular excitability efficiently.

Pathways

KCNA5 interacts with various cellular processes essential for cardiac and neuronal function. It is involved in the cardiac action potential pathway where it influences the duration and frequency of heartbeats. Moreover KCNA5 participates in the regulation of neuronal excitability through the modulation of potassium ion conductivity. It interacts with proteins such as KCNA4 and KCNA2 which also contribute to similar physiological pathways to modulate excitability.

Abnormalities in KCNA5 expression or function link to cardiac arrhythmias such as atrial fibrillation due to its role in cardiac repolarization. Additionally studies suggest a relevance to neurodegenerative disorders although research in this area is ongoing. KCNA5's relation to atrial fibrillation associates it with proteins like KCNE1 which modulate similar ion channels involved in cardiac electrical activity.

Voltage-gated potassium channel that mediates transmembrane potassium transport in excitable membranes. Forms tetrameric potassium-selective channels through which potassium ions pass in accordance with their electrochemical gradient. The channel alternates between opened and closed conformations in response to the voltage difference across the membrane. Can form functional homotetrameric channels and heterotetrameric channels that contain variable proportions of KCNA1, KCNA2, KCNA4, KCNA5, and possibly other family members as well; channel properties depend on the type of alpha subunits that are part of the channel (PubMed : 12130714). Channel properties are modulated by cytoplasmic beta subunits that regulate the subcellular location of the alpha subunits and promote rapid inactivation (PubMed : 12130714). Homotetrameric channels display rapid activation and slow inactivation (PubMed : 12130714, PubMed : 8505626). Required for normal electrical conduction including formation of the infranodal ventricular conduction system and normal action potential configuration, as a result of its interaction with XIRP2 (By similarity). May play a role in regulating the secretion of insulin in normal pancreatic islets.. Isoform 2. Exhibits a faster depolarization rate, reduced voltage-dependent recovery from inactivation and an excessive cumulative inactivation.
