Immunocytochemistry/Immunofluorescence analysis of Neuroblastoma cells (SH-SY5Y) using Anti-Kir6.1/KCNJ8 antibody [S366-60] (ab241996). Fixation : 4% PFA for 15 min. Primary Antibody : ab241996 at 1 : 100 for overnight at 4°C with slow rocking. Secondary Antibody : AlexaFluor 488 at 1 : 1000 for 1 hour at RT. 4% formaldehyde-fixed SK-N-BE (human neuroblastoma cell line) cells stained for Kir6.1/KCNJ8 (green, C) using ab241996 at 1/100 dilution for 60 minutes at room temperature in ICC/IF. Secondary antibody is Goat Anti-Mouse ATTO 488 at 1/100 dilution. Actin is detected with Phalloidin Texas Red F-Actin stain (red, B). The nuclear counterstain is DAPI (blue, A). (D) is a compsite image.

Biological function summary
Kir6.1 plays a significant role in linking cellular energy status to membrane excitability through the modulation of potassium ion conductance. It forms a complex with SUR2 (sulfonylurea receptor 2) subunits collectively constituting the K_ATP channel complex. This channel complex senses changes in intracellular ATP levels and its activity influences insulin secretion in the pancreas and vascular tone in smooth muscle. By coupling cellular metabolic status to the ion channel activity Kir6.1 contributes critically to cellular homeostasis.
Pathways
Kir6.1 channels integrate into several important biological systems. Notably the K_ATP channels participate in the cardiac muscle's ischemic conditioning pathway helping protect cells against nutrient deprivation. These channels also engage in the insulin signaling pathway affecting glucose homeostasis and energy metabolism.
