KLHL17 also known as Kelch-like protein 17 has a molecular mass of approximately 68 kDa. This protein belongs to the Kelch-like family characterized by the presence of a BTB/POZ and kelch domains. KLHL17 functions as an adaptor protein linking specific substrate proteins to the ubiquitin-proteasome system for targeted degradation. Expression of KLHL17 occurs mainly in the central nervous system.
Biological function summary

KLHL17 influences neuronal function and development. It forms part of an E3 ubiquitin ligase complex associating with Cullin 3 to facilitate protein ubiquitination. This complex plays a role in synaptic activity by regulating the levels of proteins critical for neuronal signaling. In the brain KLHL17 contributes to the fine-tuning of signaling pathways affecting cognition and memory.

Pathways

KLHL17 integrates into protein degradation and neurodevelopmental signaling pathways. It interacts with components of the ubiquitination pathway primarily linked with Cullin 3. By mediating substrate protein ubiquitination KLHL17 indirectly affects pathways like the Wnt signaling pathway essential for neuronal development. It also cooperates with proteins such as beta-catenin which participates in a wide range of cellular processes.

KLHL17 associates with neurodevelopmental and neurodegenerative diseases. Alterations in KLHL17 function may contribute to conditions such as Alzheimer's disease. Through its role in protein turnover and signaling modulation KLHL17 interacts with other proteins like amyloid precursor protein (APP) which may influence the pathological cascades associated with such disorders.

타겟 정보

Substrate-recognition component of some cullin-RING-based BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complexes. The BCR(KLHL17) complex mediates the ubiquitination and subsequent degradation of GLUR6. May play a role in the actin-based neuronal function (By similarity).
