Immunofluorescent analysis of 4% paraformaldehyde-fixed, 0.1% Triton X-100 permeabilized HUVEC (Human umbilical vein endothelial cell line) cells labeling KRIT1 with ab196025 at 1/100 dilution, followed by Goat anti-rabbit IgG (Alexa Fluor® 488) (ab150077) secondary antibody at 1/500 dilution (green). Cytoplasm staining on HUVEC cell line is observed. The nuclear counter stain is DAPI (blue). Tubulin is detected with ab7291 (anti-Tubulin mouse mAb) at 1/1000 dilution and ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution (red).
The negative controls are as follows :
-ve control 1 : ab196025 at 1/100 dilution followed by ab150120 (AlexaFluor®594 Goat anti-Mouse secondary) at 1/500 dilution.
-ve control 2 : ab7291 (anti-Tubulin mouse mAb) at 1/1000 dilution followed by ab150077 (Alexa Fluor®488 Goat Anti-Rabbit IgG H&L) at 1/500 dilution.

KRIT1 also known as Krev interaction trapped 1 and CCM1 is a protein encoded by the KRIT1 gene. This protein has a mass of approximately 99 kilodaltons. Mechanically KRIT1 functions as an adapter protein interacting with several molecular partners to regulate cellular processes. KRIT1 is expressed broadly in various tissues including in the endothelial cells of blood vessels.
Biological function summary

This protein is involved in the maintenance of vascular integrity and formation. It forms a complex with other proteins like CCM2 and PDCD10 playing a critical role in stabilizing cell-cell junctions. By organizing junctional proteins KRIT1 supports cellular architecture and mechanotransduction. This action ensures proper response to hemodynamic forces within blood vessels.

Pathways

KRIT1 is involved in the RhoA signaling pathway and integrin-dependent pathways. In the RhoA signaling pathway KRIT1 interacts with HEG1 and MAPK to facilitate signal transduction that influences cell shape and movement. Through the integrin-dependent pathway KRIT1 regulates integrins essential for cell adhesion and migration. This regulation highlights its role in mechanotransduction and vascular stability.

KRIT1 mutations associate with cerebral cavernous malformations (CCM) a condition characterized by vascular lesions in the brain. These mutations disrupt interactions with CCM2 affecting vascular development and stability. KRIT1 has connections to other cerebrovascular-related proteins indicating its involvement in leaky blood vessels that form lesions. Understanding KRIT1's interaction pathways is key to developing therapies for CCM and potentially related vascular disorders.

Component of the CCM signaling pathway which is a crucial regulator of heart and vessel formation and integrity (By similarity). Negative regulator of angiogenesis. Inhibits endothelial proliferation, apoptosis, migration, lumen formation and sprouting angiogenesis in primary endothelial cells. Promotes AKT phosphorylation in a NOTCH-dependent and independent manner, and inhibits ERK1/2 phosphorylation indirectly through activation of the DELTA-NOTCH cascade. Acts in concert with CDH5 to establish and maintain correct endothelial cell polarity and vascular lumen and these effects are mediated by recruitment and activation of the Par polarity complex and RAP1B. Required for the localization of phosphorylated PRKCZ, PARD3, TIAM1 and RAP1B to the cell junction, and cell junction stabilization. Plays a role in integrin signaling via its interaction with ITGB1BP1; this prevents the interaction between ITGB1 and ITGB1BP1. Microtubule-associated protein that binds to phosphatidylinositol 4,5-bisphosphate (PIP2)-containing membranes in a GTP-bound RAP1-dependent manner. Plays an important role in the maintenance of the intracellular reactive oxygen species (ROS) homeostasis to prevent oxidative cellular damage. Regulates the homeostasis of intracellular ROS through an antioxidant pathway involving FOXO1 and SOD2. Facilitates the down-regulation of cyclin-D1 (CCND1) levels required for cell transition from proliferative growth to quiescence by preventing the accumulation of intracellular ROS through the modulation of FOXO1 and SOD2 levels.
