Laminin subunit beta-3 (LAMB3) is a protein that is part of the laminin family commonly referred to as Laminin-332 or Laminin-5. It weighs approximately 117 kDa. LAMB3 is expressed in the epithelial basement membranes of various tissues including skin and mucous membranes. The protein forms a network essential for maintaining the structural integrity of these membranes playing a mechanical role in cellular adhesion and migration.
Biological function summary

LAMB3 functions within a heterotrimeric complex that includes Laminin-3 (alpha subunit) and Laminin-3 (gamma subunit). These interactions lead to the formation of Laminin-332 which serves as a backbone for cell attachment and spreading. It is important in processes such as epithelial cell proliferation and wound healing. By stabilizing attachments between the basement membrane and epithelial cells LAMB3 supports tissue integrity and repair mechanisms.

Pathways

LAMB3 plays a role in both the integrin signaling and epidermal growth factor receptor (EGFR) pathways. It connects with proteins like the integrin alpha-6 beta-4 complex facilitating cell adhesion and communication with the extracellular matrix. In the EGFR pathway LAMB3's interactions enhance signaling that promotes cellular proliferation and survival playing a part in skin development and maintenance.

Mutations or dysregulations in LAMB3 relate notably to junctional epidermolysis bullosa and certain cancers like squamous cell carcinoma. In junctional epidermolysis bullosa the protein loses its adhesive function affecting the epithelial junctions and causing severe skin blistering. LAMB3 also associates with integrin proteins in cancer contributing to tumor progression and metastasis when dysfunctional affecting cell adhesion and integrity.
