IHC image of LTBP1 staining in Mouse spleen formalin fixed paraffin embedded tissue section, performed on a Leica Bond^TM system using the standard protocol B. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH6, epitope retrieval solution 1) for 20 mins. The section was then incubated with ab78294, 5μg/ml, for 15 mins at room temperature. A Goat anti-Rabbit biotinylated secondary antibody was used to detect the primary, and visualized using an HRP conjugated ABC system. DAB was used as the chromogen. The section was then counterstained with haematoxylin and mounted with DPX.

For other IHC staining systems (automated and non-automated) customers should optimize variable parameters such as antigen retrieval conditions, primary antibody concentration and antibody incubation times.

Based on the immunogen sequence, this antibody is predicted to react with all three isoforms of LTBP1. The band detected here might correspond to the long isoform, which is 187 kDa and is found in fibroblasts.

LTBP1 or Latent Transforming Growth Factor Beta Binding Protein 1 mechanically acts as an important regulator in the activation and release of Transforming Growth Factor Beta (TGF-β). It is part of the latent TGF-β complex binding to TGF-β to help control its availability in the extracellular matrix. LTBP1 has a molecular weight of approximately 190 kDa. The protein is expressed in many tissues including the lung skin and heart highlighting its widespread role in various physiological processes.

Biological function summary

Its main role is modulating the bioavailability and activity of TGF-β a protein important to cellular differentiation proliferation and apoptosis. As part of a larger latent TGF-β complex it determines the precise activation and function of TGF-β by anchoring it to the extracellular matrix. This regulation is important for maintaining tissue homeostasis and has implications in developmental and repair processes across different organ systems.

Pathways

LTBP1 plays a significant role in the TGF-β signaling pathway which is important for controlling cellular growth and differentiation. It is involved in linking TGF-β to large latent complexes impacting its sequestration and activation within the matrix. Through this pathway LTBP1 interacts with proteins such as integrins which help release the active form of TGF-β furthering downstream signaling activities.
