Cell preparation containing Mouse Monocyte protein.

Specificity

This antibody is reactive to the majority of mouse monocytes and a subset of mature resident macrophages, especially those located in hemopoietic organs. Whereas all other leukocytes are negative, up to 70% of circulating monocytes stain positive with this antibody. About 30% of bone marrow cells express the antigen. Other positive macrophages can be found in the splenic red pulp, in the mesentric lymphoid paracortex, interfollicular areas of Peyer's patches and bone marrow. Few positive cells are observed in the thymic cortex and the connective tissues of the gastro-intestinal tract, the dermis and the renal medulla. Epidermal Langerhans cells also express the antigen.

This antibody can be used to identify and localize a very distinct macrophage subpopulation found in various mouse organs and its distribution pattern is unique compared to that from other macrophage-specific antibodies. The antibody is especially suitable for ontogenic studies because ER-HR3 positive macrophages are closely related to hemopoietic islands, especially at erythropoietic sites.

Macrophages and monocytes are critical components of the immune system known for their roles in phagocytosis and antigen presentation. Macrophages also referred to as tissue macrophages are derived from monocytes which circulate in the bloodstream. Monocytes with a mass of approximately 400 kDa express markers like CD14 and CD16 on their surface. They travel throughout the body via the bloodstream and infiltrate tissues where they differentiate into macrophages. Macrophages then reside in various tissues performing specialized functions based on their location such as Kupffer cells in the liver or alveolar macrophages in the lungs.
Biological function summary

Macrophages and monocytes play essential roles in the immune response and tissue homeostasis. They act as scavengers removing dead cells and pathogens through phagocytosis. Macrophages and monocytes release cytokines that regulate inflammation and recruit other immune cells to sites of infection or injury. These cells are part of a complex network involving numerous cell types and signaling molecules. Their plasticity enables them to adapt to different functional states depending on the microenvironment which is critical for balancing immune reactivity and tissue repair.

Pathways

Macrophages and monocytes are active participants in both the innate and adaptive immune pathways. They are integral to the Toll-like receptor signaling pathway which recognizes pathogen-associated molecular patterns and initiates immune responses. They also play roles in the cytokine signaling pathway interacting with proteins such as interleukin-1 and interleukin-6 to coordinate immune activities. Through these pathways monocytes and macrophages communicate with T cells and help modulate the adaptive immune response.

Macrophages and monocytes are linked to various inflammatory and autoimmune conditions. For example they are implicated in rheumatoid arthritis where they contribute to joint inflammation and damage by secreting pro-inflammatory cytokines like TNF-α. Additionally in atherosclerosis these cells participate in plaque formation within arteries by transforming into foam cells which are lipid-laden macrophages. Understanding the roles of macrophages and monocytes in these diseases helps in the development of targeted therapies highlighting their importance beyond basic immune functions.
