MCM9 also known as Minichromosome Maintenance Complex Component 9 plays a mechanical role in DNA replication. It is a component of the MCM2-7 complex which is important for the formation of the helicase that unwinds DNA during replication. MCM9 has a molecular mass of approximately 97 kDa. The expression of MCM9 is mostly found in fast-dividing cells like cancer cells as well as in tissues such as ovary and testis where cell proliferation is high.
Biological function summary

MCM9 is involved in homologous recombination repair of DNA double-strand breaks. It participates in the pre-replication complex formation and in the activation of the replication checkpoint. As part of the MCM2-7 helicase complex MCM9 maintains genomic stability by ensuring accurate DNA replication and repair. This highlights its critical role in the maintenance of DNA integrity across cell divisions particularly in proliferating cells.

Pathways

MCM9 integrates into the DNA damage response and repair mechanisms. It works alongside proteins like RAD51 a recombinase in the homologous recombination pathway for repairing double-strand breaks. MCM9 also interacts with the ATR signaling pathway which becomes activated in response to DNA damage and works to halt the cell cycle to allow repair.

Aberrations in MCM9 activity contribute to conditions such as cancer and hereditary breast and ovarian cancer syndrome. Its malfunction can lead to genomic instability and unchecked cell proliferation due to impaired DNA repair mechanisms. Connections have been drawn between MCM9 and BRCA1 another DNA repair protein highlighting its involvement in maintaining DNA stability and its association with cancer predispositions.

Component of the MCM8-MCM9 complex, which is involved in the repair of double-stranded DNA breaks (DBSs) and DNA interstrand cross-links (ICLs) by homologous recombination (HR) (PubMed : 23401855). The MCM8-MCM9 complex is a 3'-5' DNA helicase and single-stranded (ss)DNA-stimulated ATPase which binds ssDNA in the presence of nucleoside triphosphates (PubMed : 37309874). Required for DNA resection by the MRE11-RAD50-NBN/NBS1 (MRN) complex by recruiting the MRN complex to the repair site and by promoting the complex nuclease activity (PubMed : 26215093). Indirectly regulates the recruitment of downstream effector RAD51 to DNA damage sites including DBSs and ICLs, probably by regulating the localization of the MNR complex (PubMed : 23401855). Acts as a helicase in DNA mismatch repair (MMR) following DNA replication errors to unwind the mismatch containing DNA strand (PubMed : 26300262). In addition, recruits MLH1, a component of the MMR complex, to chromatin (PubMed : 26300262). The MCM8-MCM9 complex is dispensable for DNA replication and S phase progression (PubMed : 23401855). Plays a key role during gametogenesis, probably by regulating HR (By similarity).
